Cross-species single-cell spatial transcriptomic atlases of the cerebellar cortex

Recently, a paper titled "Cross-species Single-Cell Spatial Transcriptomic Atlases of the Cerebellar Cortex" performed spatial transcriptomics of the entire cerebellar cortex across two primate and one rodent species, generating a three-dimensional reconstruction of the cerebellar cytoarchitecture at single-cell resolution. This research revealed both similarities and differences between rodents and primates, contributing valuable insights into cerebellar evolution. Furthermore, it was observed that the spatial expression patterns correlated with functional connectivity within the cerebellum. These findings enhance our understanding of cerebellar functions across species. 

 

In this spatial biology-related study, researchers used STOmics Stereo-seq - the advanced single-cell spatial transcriptomics technology, which enabled the mapping of gene expression profiles with a high spatial resolution (~500 nm) and coverage of the entire cerebellum. Researchers categorized the cell types within the entire cerebellar cortex and mapped their spatial distribution in macaques, marmosets, and mice. The findings uncovered the existence of primate-specific subtypes of Purkinje cells and molecular layer interneurons (MLIs), along with the gene expression patterns enriched in specific layers and regions of the cerebellar cortex. Moreover, the researchers also noticed a correlation between the overall spatial profile of gene expression and the functional connectivity gradients of the cerebellar cortex. This was demonstrated by the resting-state fMRI in awake animals across three species. The study presents comprehensive single-cell spatial transcriptome profiles of the cerebellar cortex for macaques, marmosets, and mice.
